What is mescaline? It is the active molecule in the peyote cactus (Lophophora williamsii) of the North American deserts and the San Pedro / huachuma cactus (Echinopsis pachanoi) of the Andes, both used ceremonially for 5,000+ years. Like LSD and psilocybin it is a serotonin 5-HT2A agonist — blocking that receptor abolishes its effects — but it is the longest-lasting of the classic psychedelics: a moderate 300–500 mg dose runs ~10–12 hours, and high doses stretch toward 14. It is physiologically safe with a wide margin but reliably causes early nausea; it is US Schedule I with a religious exemption for peyote in the Native American Church, and the wild peyote it depends on is now in a genuine conservation crisis. The story of psychedelics usually starts in the twentieth century — a Swiss chemist, a bicycle, a blotter of paper. But the real beginning is older and greener than that. Long before anyone folded a molecule onto a tab, people in the deserts of what is now Mexico and Texas were eating a small, spineless cactus to speak with the sacred, and people in the Andes were brewing a tall green columnar cactus into a night-long ceremony. The compound at the center of both is mescaline, and it holds a unique place in the whole field: it is the first psychedelic humanity ever used in a documented, continuous tradition, and the first that modern chemistry ever isolated and named. The molecule: the psychedelic that started the science
Mescaline is a phenethylamine — chemically, it sits in the same structural family as dopamine and the amphetamines, which is exactly why it looks so different on paper from the tryptamines psilocybin and DMT. Its full name, 3,4,5-trimethoxyphenethylamine, describes three methoxy groups decorating a simple phenethylamine backbone. That structure matters historically: when the German chemist Arthur Heffter isolated it from peyote in 1897 and self-experimented to prove it was the active principle, mescaline became the first psychedelic ever chemically identified. Ernst Späth synthesized it in 1919, making it also the first to be produced in a lab. For half a century it was the reference psychedelic — the molecule Aldous Huxley took for The Doors of Perception, and the template Alexander Shulgin later riffed on to create an entire library of phenethylamine psychedelics. When people talk about the “classic psychedelics,” mescaline is the one that came first.
Two cacti, one alkaloid
Mescaline reaches the world through cacti, and two lineages dominate. Peyote (Lophophora williamsii) is a small, squat, spineless cactus that grows low to the ground in the Chihuahuan Desert of northern Mexico and a sliver of south Texas. Its visible crown is harvested as “buttons,” dried, and eaten; a single button carries only a modest amount of mescaline, so a ceremonial dose means many buttons. Crucially, peyote is agonizingly slow-growing — a button can take well over a decade to mature — which is the root of the conservation problem we come to below.
The other lineage is Andean. San Pedro, or huachuma (Echinopsis pachanoi, formerly Trichocereus pachanoi), and its relatives the Peruvian torch (Echinopsis peruviana) are tall, fast-growing columnar cacti from the high slopes of Peru, Ecuador, and Bolivia. The green outer flesh is sliced, boiled for hours into a bitter brew, and drunk. Because these columnar cacti grow quickly and are widely cultivated as ornamental plants, San Pedro is not endangered — a critical ecological and ethical difference from peyote. Despite its amphetamine-like skeleton, mescaline works like the other classic psychedelics. It is a serotonin receptor agonist, binding 5-HT2A (the primary driver), along with 5-HT2C, 5-HT2B, and 5-HT1A, plus some α2-adrenergic activity. The decisive evidence is pharmacological subtraction: in a controlled human study, pre-treatment with ketanserin — a selective 5-HT2A antagonist — prevented or reversed most of mescaline’s subjective effects, the same signature seen with LSD and psilocybin (Role of 5-HT2A in Mescaline, NCT04849013; Halberstadt et al., 2024). In other words, the door mescaline opens is the same 5-HT2A door on layer-5 cortical pyramidal neurons that we unpack in The 5-HT2A Receptor. Downstream, that agonism collapses the default mode network, raises neural entropy, and drives the same 5-HT2A→BDNF/TrkB plasticity pathway shared across the class. Mescaline is chemically the odd one out and mechanistically a full member of the family.
Dose and duration: the long day
What genuinely sets mescaline apart is time. The first modern dose-finding trials, run by Matthias Liechti’s Basel group, characterized pure mescaline across 100–800 mg in healthy volunteers. Effects come on slowly — often over one to two hours, frequently accompanied by early nausea — and then last a very long time: mean duration rose from about 6.4 hours at 100 mg to roughly 14 hours at 800 mg, with a moderate 300–500 mg dose landing near 10–12 hours (Ley et al., Translational Psychiatry 2024). That makes mescaline the longest-acting of the classic psychedelics — longer than LSD’s ~9–12 hours and roughly double psilocybin’s ~4–6. In a direct comparison of mescaline, LSD, and psilocybin, the Basel team found the three altered states were qualitatively similar at matched intensity — once again pointing to the shared 5-HT2A mechanism — with duration the standout difference. In cactus form the math compounds: a full peyote dose can mean five or more buttons (on the order of 25–50 mg mescaline each), and a San Pedro brew is dosed by the dried gram — both delivering a long, bitter, physically demanding onset before the visionary phase arrives.

Phenomenologically, mescaline is often described as warm, richly visual, and emotionally open — heavy on flowing geometric and color imagery, with a notably embodied, “earthy” character that many find gentler and more relational than LSD’s electric edge. But the honest caveat is the same one that applies across this class: rigorous controlled data emphasize similarity among the classic psychedelics once dose and duration are matched, and the “vibe” contrasts are widely reported tendencies rather than firmly quantified facts. Physiologically, mescaline is a mild sympathomimetic: the Liechti safety-pharmacology work found it raised blood pressure and heart rate dose-dependently, with nausea and vomiting the most common adverse effects at the 800 mg dose (Klaiber et al., Br. J. Clin. Pharmacol. 2024). The nausea is not incidental — in traditional peyote and huachuma ceremonies the purge is often understood as part of the medicine itself.
The sacred lineage: a sacrament that outlasted prohibition
Mescaline’s ceremonial history is the deepest of any psychedelic. Peyote use in the Americas is chemically confirmed to be at least ~5,700 years old (radiocarbon-dated buttons from Texas), and San Pedro is depicted in Andean art at Chavín de Huántar in Peru going back roughly three thousand years (Society of Ethnobiology). In the North, the peyote tradition crystallized into the Native American Church (NAC), formally chartered in 1918, which blends Indigenous peyote ceremony with elements of Christianity and today counts hundreds of thousands of members across the U.S. and Canada. The Wixárika (Huichol) of Mexico still undertake an annual pilgrimage to Wirikuta, their sacred peyote desert in San Luis Potosí. In the Andes, huachuma remains central to curanderismo healing ceremonies. This is not a recreational drug that acquired a spiritual veneer; it is a living sacrament with an unbroken lineage — a fact that shapes both the law and the ethics around it.
The peyote crisis: the plant itself is running out
Here is the part of the mescaline story most people never hear. Wild peyote is in serious decline. Because the cactus grows so slowly and reproduces so gradually, it cannot keep pace with harvesting pressure — and that pressure has multiplied. The Native American Church declared a “peyote crisis” a top priority back in 1995, and the situation has worsened since: root-plowing and land development (including agriculture and energy projects) across the Texas peyote gardens, illegal poaching, improper harvesting that kills the root, climate stress, and surging demand driven by psychedelic tourism and worldwide internet sales (KUER, 2024). The response from Native communities — through efforts like the Indigenous Peyote Conservation Initiative — has a clear message that surprises many decriminalization advocates: most peyote people want the plant kept out of the psychedelic gold rush entirely, protected and reserved for Indigenous ceremonial use, precisely because broad decriminalization would accelerate its extinction in the wild. This is why the sustainable, fast-growing San Pedro matters so much as the ecological alternative for the mescaline-curious: choosing the columnar cactus over the button is, increasingly, a conservation act.

Like the other classic psychedelics, mescaline has a wide physiological safety margin and no established lethal overdose from its direct pharmacology at typical doses; it is not physically addictive, and it produces rapid tolerance. The reliable physical events are the nausea and vomiting of onset and a modest, transient rise in heart rate and blood pressure — meaning people with significant cardiovascular disease should be cautious. The dominant real risk, as with all psychedelics, is psychological: mescaline’s exceptionally long duration means a challenging experience can unfold over ten hours or more, a longer window than any other classic psychedelic and a heavier demand on set, setting, and support. A personal or family history of psychosis is a standard contraindication, and combining mescaline with other serotonergic drugs (including MAOIs and many antidepressants) is a real interaction concern. Legality in 2026: Schedule I, with a sacred exception
Federally, mescaline is Schedule I in the United States — and so is peyote specifically. The major exception is religious: under the American Indian Religious Freedom Act Amendments of 1994 (and DEA regulation 21 CFR 1307.31), the sacramental use of peyote by enrolled members of the Native American Church is federally protected. Outside that framework, possession is illegal under federal law. At the state level, Colorado’s Proposition 122 (2022) is notable: it decriminalized personal use of several natural psychedelics including mescaline — but the measure deliberately excludes peyote, explicitly to protect the vulnerable cactus and honor Indigenous stewardship. Live San Pedro cacti occupy a strange legal niche: the plants are widely and legally sold as ornamentals, but extracting or consuming their mescaline is not lawful. An April 2026 federal executive order directed agencies to accelerate psychedelic research — naming several compounds — but did not reschedule or legalize mescaline. The honest bottom line
Mescaline is the psychedelic that started it all — the first isolated, the longest used, and in many ways the most humbling. Pharmacologically it is a full member of the classic-psychedelic family, opening the same 5-HT2A door as LSD and psilocybin and producing a qualitatively similar altered state; what sets it apart is the length of the day it asks for and the nausea it charges at the gate. But mescaline’s most important lesson is not chemical. It is that a psychedelic can be a living sacrament with a five-thousand-year lineage and an endangered body — that the responsible path forward means honoring Indigenous stewardship, protecting the wild peyote gardens, and, for the curious, looking to the sustainable San Pedro rather than the vanishing button. The oldest psychedelic still has the most to teach about how to treat these medicines with respect.
